Look again at the results
obtained following isolation of the food vacuoles at different time points
and examining the existence of proton pumps using various antibodies. (A.
Check
procedure, B. Check original
results.)
1. Assume that no bands
are detected in lane 2 (purified proton pump), but
the same band as in the original results is detected in lane 7 (lysate
of the whole Paramecium reacted only with the secondary antibody).
Please draw what bands you expect to see in each of all the other lanes
and explain your drawing.
2.In view of these new results, can you draw any conclusions from this experiment? Explain. |

3. Assume
that in lane 7 (lysate of the whole Paramecium reacted
only with the secondary antibody) you observe one
band of the same size as that of the purified proton pump (56.1 Kd) observed
in lane 2 (purified proton pump).
Please draw what bands you expect to see in each of the other lanes
and explain your drawing.
4. In view of these new results, can you draw any conclusions from this experiment? Explain. |
|
5. Assume that lane 4 (lysate
of the Parameciun food vacuole collected 0.5 min after feeding)
contains the same bands as lane 5 (lysate of the Parameciun
food vacuole collected 5 min after feeding). Please
explain these results.
6. Do these new findings support any of the previously described hypotheses (B or C)? 7. In view of these findings, should you still check wether addition of NEM affects any of the food vacuole features which change with time? Explain. (Check procedure ) 8. Describe the results you expect to find, if you perform this experiment. |
