Remember that the arbitrary start point of pGEM is at the beginning of the T7 promoter, and the  numbering of the restriction sites goes from 14 (ApaI) to 109 (NsiI). Notice that GOI is inserted in the reverse orientation (like the reporter gene lacZ and the f1 origin). When inserted like that GOI is under the control of the SP6 promoter.
